How to get to Dawei:

Flights from Yangon:

Myanmar National Airline and Air KBZ offer daily flights to Dawei; and Golden Myanmar airlines flies 3 days (TUE/THU/SAT) per week. 

Busses from Yangon: 

Several different companies operate buses, which leave Yangon in the evening and arrive in Dawei early the following morning.

Train from Yangon:

The train takes approximately 24 hours from Yangon.

Train from Ye:

By Car From Yangon:

Drive to Tanintharyi from Yangon through Bago, Kyaikto, Mawlamyine, Mudon and Yevia Mawlamyine and Ye.
It's about a 13-to-16-hour drive.  

How to get to Myeik:

Flying to Myeik:

Airlines flying from Yangon to Myeik include Myanmar Airways International,Air KBZ,Myanmar National Airlines.( Flight Time:1:35hr ).
Airlines flying from Dawei to Myeik ( 45 min ).

By Bus or Car to Myeik:

From Yangon by bus or by car( 18hr )
From Dawei by bus or by car ( 6hr )
From Kawthaung by bus or by car ( 7hr 20min ) .

How to get to Kawthaung: 

From Yangon to Kawthaung with AIR KBZ and Golden Myanmar Airline. (Flight Time: 2hr40min+) 
from Dawei to Kawthaung 1hr15min by Myanmar National Airlines and KBZ flight

By car or by bus:
From Yangon to Kawthaung - 1280 km (26hr8min).
From Dawei to Kawthaung 670 km (13hr18min).

By Boat:

From Ranong Thailand - (about 25 minutes)

To Tanintharyi from Thailand:

It is possible to travel overland from Thailand, via the Phu Nam Ron / Tikki border crossing. 

Note that currently, international travelers using the Phu Nam Ron / Tikki border crossing must have a visa from a Myanmar embassy to make this crossing. E-visa are not currently accepted due to unreliable internet connections.
